摘要 目的观察蒺藜皂苷(GSTT)对血管平滑肌细胞(VSMC)增殖及相关因素的影响。方法采用培养的牛VSMC,以血管紧张素Ⅱ(AngⅡ)为刺激因子,应用MTT法检测细胞增殖;以H2O2为刺激因子,应用激光扫描共聚焦显微镜检测VSMC内钙离子浓度;应用硝酸还原酶法检测培养的牛血管内皮细胞NO释放量。结果GSTT能明显抑制AngⅡ刺激的VSMC增殖,具有剂量依赖性;GSTT也能抑制H2O2所致的VSMC内钙离子浓度升高,使血管内皮细胞释放的NO明显增多。结论GSTT能抑制AngⅡ诱导的VSMC增殖,此作用可能与抑制活性氧激活钙离子信号转导途径、增加血管内皮细胞NO含量等因素有关。 OBJECTIVE To investigate the effects of gross saponins from tribulus terrestris (GSTT) on the proliferation of vascu- lar smooth muscle cells (VSMC). METHODS The bovine VSMC were cultured and the effect of GSTT on cellular proliferation in VSMC stimulated by angiotension Ⅱ ( Ang Ⅱ ) were assayed by MTT method, and the effect of GSTT on intracellular free calcium ( [ Ca^2+ ]i)in VSMC stimulated by H2O2 were also assayed by laser scanning confocal microscope. The release of nitric oxide(NO) in bovine vascular endothelia cell were detected by the method of nitric acid reductase. RESULTS GSTT could significantly inhibit the proliferation of VSMC stimulated by Ang Ⅱ , which has the dose-dependent effects. GSTT also shown to be able to depress increase of [ Ca^2 +] i stimulated by H2O2 , and increase the release of NO. CONCLUSION The inhibition effect of GSTT on the proliferation of VSMC stimulated by Ang Ⅱ may be related with the suppression of reactive oxygen activated Ca^2 + signal path and increase the release of NO.
