小儿止咳平喘露的平喘作用及其对外周血可溶性细胞黏附分子-1的影响 被引量:1

摘要 目的在小儿止咳平喘露平喘、镇咳、祛痰的实验基础上,观察其对哮喘动物模型肺泡灌洗液嗜酸性粒细胞计数及外周血可溶性细胞黏附分子-1(sICAM-1)的影响。方法①将预选合格的豚鼠30只随机分为3组,即正常对照组,细辛脑组,小儿止咳平喘露组,每组10只。采用豚鼠组胺引喘法观察小儿止咳平喘露的平喘作用。②采用豚鼠枸橼酸引咳法观察小儿止咳平喘露的镇咳作用,给药方式及剂量同平喘试验。③将ICR健康小鼠30只,随机分为3组,分组方式同平喘试验,采用小鼠气道酚红排泄法观察小儿止咳平喘露的祛痰作用。④将40只健康豚鼠随机分为4组,即正常对照组、模型组、细辛脑组、小儿止咳平喘露组,每组10只。建立卵蛋白致敏豚鼠哮喘模型,在肺泡灌洗液中计数白细胞总数和嗜酸性粒细胞数,并用ELISA法检测豚鼠血清sICAM-1的表达水平。结果小儿止咳平喘露能明显延长组胺诱导的豚鼠引喘潜伏期和枸橼酸引起的豚鼠咳嗽潜伏期,促进小鼠气道酚红排泄,降低哮喘豚鼠肺泡灌洗液中嗜酸性粒细胞计数和血清sICAM-1水平。结论小儿止咳平喘露具有平喘、止咳、祛痰作用,其作用机制可能与降低哮喘豚鼠肺泡灌洗液中嗜酸性粒细胞计数和血清sICAM-1水平有关。 OBJECTIVE To evaluate the antiasthmatic effects of Zhikepingchuan Decoction For Children and its effect on blood sICAM-1 level. METHODS (1)Antiasthmatic effect was investigated with asthma induced by histamine aerosol in guinea pigs. (2)Antitussive effect was studied with citric acid-induced cough in guinea pigs. (3)Expectorant effect was studied with excretion of phenol red in the airway of mice. (4)Forty guinea pigs were randomly divided into four groups : control, model, α-asarone, Zhikepingchuan Decoction For Children ,with 10 guinea pigs in each group. Experimental animals were sensitized and challenged by OVA to establish an asthmatic guinea pig model. The numbers of leukocytes and eosinophils( EOS )in the bronchoalveolar lavage fluid (BALF) were detected,and the levels of plasma slCAM-1 in four groups were determined respectively by enzyme-linked immunosorbentassay (ELISA). RESULTS Zhikepingchuan Decoction For Children can obviously prolong the incubation times both for histamine-induced asthma and for citric acid-induced cough ; It can also increase the excretion of phenol red and reduce either the number of EOS in BALF or the level of slCAM-1 in plasma. CONCLUSION These results indicate that Zhikepingchuan Decoction For Children has some pharmacological effects of antiasthma, antitussive and expectorant. The mechanism of these effects might be associated with reducing the number of EOS in airway and/or declining plasma sICAM-1 level.
