香丹制剂对LPS诱导的血管内皮细胞NF-κΒ活性的影响 被引量:4

摘要 目的:探讨中药复方香丹注射液对血管内皮细胞核转录因子NF-κB及抑制因子IκB活性的干预作用。方法:采用MTT法及形态学方法观察香丹注射液对人脐静脉血管内皮细胞活性的影响,用Sandwich ELISA定量分析法和定量免疫细胞技术,检测NF-κB活性及胞浆内IκB含量。结果:有效剂量范围的香丹注射液能抑制血管内皮细胞的生长,拮抗NF-κB活性及抑制IκB降解(P<0.01)。结论:香丹注射液的抗炎作用可能与其拮抗NF-KBp65活化有关。 Objective: To examine the effect of the traditional Chinese drug Compound Xiangdan injection (XDI) on the LPS-mediated NF-κB activation in human umbilical vein endothelial cells (HUVECs) .Methods: The effects of XDI on the growth of HUVECs were observed by MTr and morphological method.The amounts of cytoplasm IkB and nuclear NF-κB in ECV-304 cells with or without LPS treatment were determined by the quantitative immunocyte assay and Sandwich ELISA quantitative assay. Results: XDI had some inhibitory effect on the growth of ECV304, the activatiojn in the nuclear NF-κB and the IkB reduction of cytoplasm (P 〈 0.01 ) . Conclusion: The anti-inflammation of XDI may be correlated with the down-regulation NF-κB activation in the human vascular endothelial cells.
