摘要
目的探讨红景天对新生大鼠缺氧缺血性脑病(HIE)的疗效及可能的作用机制。方法36只7日龄新生SD大鼠随机分为假手术组、实验组和中药组,手术分离左侧颈总动脉,假手术组不再做任何处理;实验组和中药组结扎并切断左侧颈总动脉,2h后置于8%氧氮混合气体2h。中药组于模型建立后2h开始灌喂红景天,每日1次,每次每只1ml。连续5d,休息2d再灌喂5d。分别于实验第1日和第3日处死动物,留取脑组织,半数中性福尔马林固定,半数液氮保存,采用HE染色、RT-PCR和免疫组化,在蛋白和mRNA水平检测新生大鼠HIE时HIF-1α基因的表达。结果缺氧缺血后大鼠相继表现为烦躁不安、全身发绀、呼吸加深加快、站立不稳、嗜睡、激惹或间断发作的痉挛和抽搐;HE染色显示实验组大鼠左侧大脑皮层出现局灶性神经元核固缩,核碎裂,核仁偏位,不清或消失,基质疏松;中药组大鼠脑组织结构基本接近假手术组,无明显水肿和坏死表现;实验组和中药组大鼠脑组织HIF-1α mRNA(RT-PCR)表达较假手术组明显增加,尤其是中药组实验第3日者;免疫组化显示实验组和中药组各时间点大鼠大脑皮层和海马区均可见不同程度的HIF-1α表达,明显高于假手术组,阳性表达主要在血管内皮细胞,海马和皮层的锥体细胞亦有HIF-1α阳性细胞分布。结论红景天对新生大鼠HIE有一定的治疗作用,其可能的作用机制之一是增加脑组织HIF-1α的表达。
Objective: To get the message of expression of chinese traditional medicine Hongjingtian detect on neonatal rats with hypoxic -ischemic encephalopathy(HIE), so to find out the more effective method and mechanism to treat HIE. Methods: 36 neonatal rats aged 7d were divided randomly into three groups, sham group, experimental group and chinese traditional medicine group with 12 rats in each. The left common carotid artery of the neonatal rats in experimental group and chinese traditional medicine group was ligated. 2h later, they were subjected to inhalation of 8% oxygen for 2h. After 2h chinese traditional medicine group was fed Hongjingtian, each rat with 1ml every time, qd. Continus 5d, rest 2d againe feeding 5d. According to the time point, 12 rats of every group were further divided randomly into two groups including ld and 3d, with 6 rats in each subgroup. The expression of HIF - 1α in the levels of protein and mRNA in braintissues of the rats were determined with HE staining, immunohistochemical staining and RT - PCR. Results: Neonatal rats in hypoxic - ischemicgroup behaved restlessness, cyanosis, deep and rapid breath, astasia, lethargy, irritation and spasm; There were focal pyknosis, fragmentation, dissymmetry, blur or disappearance of nucleoli, raritas of ground substance in left cerebral cortex; but there was no statistic significance in chinese traditional medicine group and sham group. The expression of HIF - 1α mRNA in brain tissue of experimental group and chinese traditional medicine group were higher than that in sham group, especially chinese traditional medicine group at 3d; HIF- 1α positive cells were found in cerebral cortex and hippocampus. There were more HIF- 1α positive cells in experimental group and chinese traditional medicine group and mainly in vascular endothelial cells. Conclusion: Chinese traditional medicine Hongjingtian have some effect on HIE rats, it is likely to increase the expression of HIF- 1α in brain tissue.
