摘要
目的:建立适合卡马西平、苯巴比妥、苯妥英钠三种抗癫痫药物系统分离的胶束电动毛细管色谱(MECC)方法。方法:考察了运行缓冲液体系中各种参数,包括SDS浓度、电泳缓冲液浓度、pH值、分离电压对电泳分离效果的影响;MECC优化分离系统为:石英毛细管75μm×37cm,电泳缓冲液为15mmol/L磷酸缓冲液(pH=9.4)含十二烷基硫酸钠(SDS)25mmol/L。分离温度20℃,分离电压25KV,压力进样:10psi×5s,正极进样,负极柱上200nm检测。结果:三种常用抗癫痫药物卡马西平、苯巴比妥、苯妥英钠在5分钟之内获得了理想的分离。结论:胶束电动毛细管色谱法可以快速有效地分离和检测上述三种抗癫痫药物。
Objective , to establish a capillary electrophoresis method for system separation of three anti-epilepsy drugs (phenytoin, carbamazapine and phenobarbital) based on the micellar etectrokinetic caPillary chromatography (MECC) mode. Methods : The optimum separation conditions were : capillary 75 μm × 37 cm uncoated; running Puffer pH 9.4, background electrolyte (25 mmol/L SDS, 15 mmol/L disodium hydrogen Phosphate); applied voltage 25 KV (+) (-); injection 10 psi × 5 s ; on-column detection 200 nm; column temlaerature 20 ℃. Results, The ideal separation for phenytoin, carbamazapine and phenobarbital was obtained within 5min. Conclusion ; The mioellar electrokinetic capillary chromatography is an effective and fast way to separate the three anti-epilepsy drugs.
