Determination of Platelet Microparticles in Peripheral Blood from Patients with Idiopathic Thrombocytopenic Purpura by Flow Cytometry

摘要 目的建立CD61与Annexin V双染法检测外周血中循环血小板微粒的方法,探讨血小板微粒在特发性血小板减少性紫癜患者中的临床意义。方法使用流式细胞术检测了12例特发性血小板减少性紫癜患者和15例健康对照者外周血中血小板微粒的含量。采用CD61与Annexin V双染法,CD61弱阳性并且Annexin V强阳性者定义为血小板微粒,分析血小板微粒的百分含量。结果CD61与Annexin V双染法能够清楚地区分出富血小板血浆中血小板微粒。特发性血小板减少性紫癜患者血小板微粒为12.3%±5.4%,健康对照组为1.1%±0.3%,P<0.05,差异显著。结论CD61与Annexin V双染法能够准确检出外周血中循环的血小板微粒。血小板微粒检测在特发性血小板减少性紫癜患者中有一定的临床意义。 Objective To establish a method of detecting platelet microparticles (PMPs) in platelet-rich plasma by flow cytometry (FCM) ,and evaluate its clinical significance in patients with idiopathic thrombocytopenic purpura (ITP). Methods Detected the PMPs with Annexin V and CD61 by FCM. Co-expression of Annexin V and weakly CD61 were considered PMPs,PMPs were examined in 12 patients with ITP and control groups. Results PMPs were identified clearly with Annexin V and CD61 from platelet rich plasma. The PMPs in patients with ITP were 12.3%±5.4%,it was more higher than control groups (1. 1% ± 0. 3%, P 〈 0. 05). Conclusion The method is established for determination of PMPs in platelet-rich plasma by two color FCM. More higher level of PMPs in patients with ITP was found.
