大肠杆菌脂多糖和氢化考的松诱导胸腺细胞凋亡的实验研究 被引量:1

摘要 用琼脂糖凝胶电泳、电镜等方法研究大肠杆菌脂多糖(LPS)和氢化考的松所致的小鼠胸腺细胞凋亡,以建立适于实验的细胞凋亡模型,并确定了凋亡模型的最佳用药剂量和时相。实验结果表明,LPS和氢化考的松均可诱导小鼠胸腺细胞发生凋亡,LPS腹腔注射量为25μg/只时造模较为适宜,造模最佳时相为腹腔注射18h,氢化考的松模型经测定其最佳时相为肌肉注射24h。两种凋亡模型比较,LPS模型造模时间短(仅需18h),简便易行,是一种较好的研究细胞凋亡的实验工具。 Apoptosis is at the cutting edge of histomorphology and biochemisry. This paper describes the attempt to find and apoptosis model suitable for the research of traditional chinese medicine. Two mouse thymic apoptotic models induced by lipopolysaccharide (LPS) or hydrocortisone were studied. The experiment showed the best medical dose and time course of these two models by agarose gel electrophoresis and electronic microscope observation. the results indicated that LPS or hydrocortisone could induce apoptosis in mouse thymus. In LPS model, the dose of LPS 25μg per mouse injected intraperitoneally(I.P) was most appropriate and 18 hours after intraperitoneally (I>P) was the best time course. The best time of hydrocortisone model was 24 hours intramuscularly(I.M). For different virtues of these two models, LPS model have some advantages than hydrocortisone, such as short time to establish (only 18 hours), so it is more suitable for the experimental research of apoptosis in mouse thymus.
