大鼠自体静脉移植后内膜增生与平滑肌细胞增值 被引量:6

DYNAMIC CHANGES OF INTIMAL HYPERPLASIA AND PROLIFERATION OF SMOOTH MUSLE CELL AFTER AUTOGENOUS VEIN-TO-ARTERY GRAFTING IN A RAT EXPERIMENTAL MODEL

摘要 研究大鼠颈外静脉移植入颈总动脉后内膜增生(IH)的发病机制。应用组织切片的特殊染色,单克隆抗PCNA抗体及抗Dm抗体的免疫组化方法观察IH的发生过程。结果:①术后1d移植静脉的内膜消失,术后2d出现IH并逐渐增生,IH中可见大量平滑肌细胞(SMC),术后12周IH的面积与术后18周元显著差异;②IH中SMC的Dm呈阳性;③IH中SMC的PCNA指数于术后2d骤然升高,术后2周达高峰。结论:IH发生在移植术后2d,逐渐增厚至术后12周方停止,术后早期发生的IH是由中膜的SMC移行至内膜并增殖所致。SMC增殖的高峰在术后第2周。 PURPOSE To study the relationship between the dynamic changes of intimal hyperplasia (IH) and proliferation of smooth muscle cell (SMC) after autogenous vein - to - artery grafting in rats.METHODS The autogenous jugulaLr vein were grafted into the carotid artery. The histological cross section of vein grafts were stained with Van Gieson and weigert stain for morphometric evaluation. PCNA and Dinwere immunohistochemi call y locallized in histological cross section of vein grafts.RESULTS IH of vein graft began to appear at 2 days after grafting and then to increase gradually. ManySMC in IH were observed. The enointimal cross - sectional area at 12 weeks and 18 weeks after grafting werenot significant difference. There were expression of Dm in IH. The PCNA index of SMC in IH rose abruptly at2 days after grafting and reach a peak at 2 weeks.CONCLUSIONS IH of vein graft arises at 2 days after grafting and reach maximum at 12 weeks. Hereafter it remained steady. IH is the result of migration and proliferation of SMC from the media into the intima atearly stage and doposition of extracellular matrix (ECM) in late stage after vein grafing. The SMC proliferationreaches a peak at 2 weeks.
