摘要 用硫酸铵分段盐析、DEAE纤维素-DE52离子交换柱层析、SephadexG100凝胶过滤柱层析等方法.从蚯蚓组织中提取有促髓系幼稚细胞增殖活性的蛋白质,提纯倍数达190倍。用聚丙烯酰胺凝胶电泳检测行5条区带;蛋白酶复印结果显示其中3条具有蛋白酶活性。该活性蛋白质有一定耐热性(55C,60min),促增殖活性强的部分其纤清活性也强。提示该活性蛋白属于蛋白酶,可能是细胞增殖信息传递链的组分或是使传递链组分活化的物质。 By means of ammonium sulfate precipitation, ion exchange chromatography (DEAE cellulose DE52) and gel filtration chromatography (Sephadex G 100), some proteins with proliferation improving aetivity on myelocytic cell were extracted from earthworm. The enrichment ratio was 190.There were five bands in polyacrylamide gel electrophoresis (PAGE) and three of the bands showed fibrinolytic activity in zymography. The active proteins were heat-resistant (55C, 60 min ). It was showed that the fraction with righter proliferation improving activity had higher fibrinolytic activity as well. It suggests that the active proteins may be proteases, and they may be the constituents of cell proliferative information transport ation chain or the activators of the constituents.
