B7-1阳性细胞的选择与鉴定

SELECTION AND IDENTIFICATION OF B7-1 POSITIVE CELLS

目的选择和鉴定共刺激分子Ｂ７—１阳性的细胞。方法应用Ｇ４１８进行选择培养，然后用荧光激活的流式细胞计数仪（ＦＡＣＳ）进行鉴定，结果Ｇ４１８选择前，阳性细胞率为３７．２５％，应用２００μｇ／ｍｌ４００μｇ／ｍｌ的Ｇ４１８选择后，阳性细胞率分别为５２％和８８．８８％。结论Ｇ４１８浓度高时，选择效果较好；ＦＡＣＳ是一种快速而且敏感的鉴定方法。

Objective To select and identify B7-1 positive cells. Methods The cells were selected with G418,and then identified with Fluorescein Activation Cell Sorter(FACS).Result Before the use of G418,the rate of positive cells was 37.25% and after selection with 200g/ml and 400g/ml G418,the rates of positive cells were 52% and 88.88%.Conclusion When G418 has a high concentration,the result of the selection is better.FACS is a quick and sensitive method of identification.
