缺氧因子在人鼻黏膜上皮细胞的表达研究 被引量:5

摘要 目的探讨在感染性因子、炎性因子的作用下人鼻黏膜上皮细胞缺氧诱导因子-1α(HIF-1α)的表达情况。方法无血清原代培养人鼻息肉及下鼻甲黏膜上皮细胞,以脂多糖(LPS)10,100,1000ng/ml,白细胞介素-1β(IL-1β)20ng/ml作用于对数生长期的上皮细胞3,6,9h后,采用免疫细胞化学方法和原位杂交方法检测HIF-1α蛋白及mRNA的表达情况。结果①鼻黏膜上皮细胞在感染性因子LPS作用下,HIF-1α表达增加并且具有时间依赖性和计量依赖性,其中以100ng/ml,6h表达最明显(P<0.05);②鼻黏膜上皮细胞在炎性因子IL-1β的作用下,HIF-1α表达趋势同LPS组,表达强度略低于LPS100ng/ml,但二者无显著性差异;③各组因素作用下鼻息肉组HIF-1α的表达明显高于下鼻甲组(P<0.05)。结论感染性因子、炎性因子作用下鼻黏膜上皮细胞表达HIF-1α增加,进而诱导相关因子的大量表达,是鼻息肉发生发展的重要原因之一。 Objective To observe the expression and the role of hypoxic inducible factor ( HIF)-1α induced by inflammatory factor and infection factor in primary human nasal epithelial cell (HNEC). Methods Epithelial ceils of nasal polyps (NP) and inferior turbinate (IT) were cultured without serum under stimulus of LPS 10,100, 1 000 ng/ml and IL-1 β 20 ng/ml for 3,6,9 h respectively. The expression of HIF-1α protein and mRNA derived from epithelial ceils was detected by immunocytochemistry and in situ hybridization. Results (1) The expression of HIF-1α increased under the stimulus of inflammatory factor with the increasing of time and concentration, especially by LPS 100 ng/ml at 6 hour( P 〈 0. 05 ) ;(2)The expression tendency of HIF-1α induced by IL-1 β was the same as that by LPS groups, and the expression intensity of HIF-1α induced by IL-1β was lower than that by LPS 100 ng/ml, but there was no differences between them. (3)The HIF-1α expressed more intensively in epithelial ceils from NP than IT under all conditions ( P 〈 0. 05 ) ; Conclusions Epithelial ceils actively produce vast HIF-1α under inflammatory factor and infection factor to induce overexpressed correlated factors, which is one of important factors of formarion of NP.
