格尔德霉素对肝细胞生长因子引起的人脑胶质瘤细胞增殖与凋亡变化的影响 被引量:5

摘要 目的研究格尔德霉素(GDM)对肝细胞生长因子(HGF)引起的胶质瘤细胞增殖、细胞周期和细胞凋亡的影响。方法U251-MG和U87-MG细胞同步生长后,用HGF作用24h,加入不同剂量GDM(终浓度为50、250、500与1000nmol/L)再处理48h。MTT法检测细胞增殖能力,流式细胞术检测细胞周期时相和细胞凋亡变化。结果与正常对照组相比,单纯HGF组其U251-MG和U87-MG细胞增殖指数显著增高(P<0.05),凋亡百分率显著降低(P<0.05);单纯GDM组这两种肿瘤细胞增殖指数显著低于正常细胞(P<0.05),凋亡细胞百分率显著高于正常组(P<0.05),且随浓度增加其增殖抑制作用逐渐增强,1000nmol/L的GDM对HGF的抑制效果最好;HGF联合不同浓度GDM组其增殖指数则显著高于单纯GDM组,凋亡细胞百分率显著低于GDM组;同时HGF联合不同浓度GDM组其细胞G0/G1期细胞表达百分率显著高于GDM组(P<0.05),而G2/M期细胞表达百分率显著低于GDM组(P<0.05)。结论GDM能抑制HGF诱导的胶质瘤细胞的增殖,并促进其凋亡,即GDM对HGF的增殖促进及凋亡抑制作用具有逆转能力。
