摘要
目的:探讨猫爪草皂苷对人乳腺癌MCF-7细胞生长的抑制作用及对免疫功能的影响。方法:四唑盐比色法、集落形成实验测定猫爪草皂苷对体外培养的MCF-7细胞生长的抑制率;DNA Ladder、流式细胞术(FCM)检测细胞凋亡;不同方法测定猫爪草皂苷对正常小鼠免疫功能的影响。结果:猫爪草皂苷抑制MCF-7细胞生长和集落形成,呈量-效关系;DNA Ladder、FCM显示其可有效诱导细胞凋亡;猫爪草皂苷灌胃给药(2.96,5.93,11.86g生药.kg-1)明显升高雌性小鼠脾指数和淋巴细胞转化率,增加NK细胞活性。结论:猫爪草皂苷可提高正常小鼠免疫调节功能,明显抑制MCF-7增殖诱导其凋亡。
OBJECTIVE To study the effect of extracts in Radix Ranunculi Ternati on anti-tumor of MCF-7 cells in vitro. METHODS MCF-7 human breast carcinoma cell lines were treated with the extracts in Radix Ranunculi Ternati in vitro. Cell proliferation was evaluated by MTT assay; the conlony forming was applied in demonstrating effect on anti-tumor of MCF-7 with different concentration; and the rate of apoptosis and distribution of cell cycle were determined by DNA Ladder and flow cytometry. The experiments of the immune function were studied. RESULTS The MCF-7 cells in vitro experiment multiplies the inhibitory action lengthened along with the density of the Radix Ranunculi Ternati withdraws enhancement and the response time but is brighter. The experiment of the proliferation of lymphocyte and NK cell activities reveals also the extracts strengthens the non-special immune function of the normal mice. CONCLUSION The Radix Ranunculi Ternati withdraws inhibits MCF-7 cells growth via apoptosis reduction and the extracts strengthens the non-special immune function of the normal mice.
