摘要
目的:探讨天麻素对缺血性脑损伤的神经保护作用.方法:通过线栓技术闭塞大脑中动脉(MCAO)2h诱导建立暂时性局部脑缺血模型,SD大鼠腹腔内连续注射天麻素3d.采用TUNEL染色测定缺血边缘区细胞凋亡;免疫组织化学染色观测S100β和iNOS蛋白表达.结果:MCAO后3d,天麻素组大鼠脑梗死边缘区TUNAL+细胞数目明显减少(P<0.05);S100β和iNOS+细胞数目下降(P<0.05).结论:天麻素可以抑制脑缺血诱导的细胞凋亡,具有神经保护作用;S100β和iNOS蛋白表达的下调可能涉及到天麻素的抗细胞凋亡作用.
AIM: To assess the neuroprotective action of gastrodin on ischemic brain injury in vivo. METHODS: Gastrodin was intraperitoneally injected daily for 3 consecutive days starting 2 h after focal cerebral ischemia induction in SD rats by intraluminal occlusion of middle cerebral artery(MCAO). Apoptosis in the ischemic boundary zone was evaluated with TUNEL staining. Immunohistochemical techniques were used to analyze the S100β and iNOS expressions. RESULTS: Three days after MCAO, the number of TUNEL positive cells in gastrodin group was significantly decreased in the ischemic boundary zone(P 〈0.05) , and the numbers of S100β and iNOS positive cells were remarkably decreased as well ( P 〈 0. 05 ). CONCLUSION: Gastredin injection could depress the apoptosis in a rat model of focal cerebral ischemia, suggesting that gastrodin has a neuroprotective effect on brain ischemia. Down-regulation of S100β and iNOS expressions is possibly associated with the inhibitory effect of gastrodin on apoptosis under ischemic conditions.
