肾素血管紧张素Ⅱ-1型受体基因多态性与2型糖尿病伴或不伴有高血压的相关性 被引量:2

摘要 目的:分析宁夏地区人群肾素血管紧张素Ⅱ-1型受体基因(AGTR1)A10208G位点多态性与2型糖尿病及2型糖尿病合并高血压之间的关系.方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术分析85例单纯2型糖尿病患者(DM组)、42例2型糖尿病合并高血压患者(DM-H组)及224例健康对照者(NC组)AGTR1基因A10208G位点多态性,并测定其相应生化指标.结果:DM组AGTR1基因A10208G位点基因型分布和等位基因频率与NC组比较差异有统计学意义(基因型:χ2=5.94,P<0.05;等位基因:χ2=4.03,P<0.05);DM-H组携G等位基因患者BMI、收缩压、舒张压均显著高于DM组携G等位基因患者,差异有统计学意义(P<0.01).结论:AGTR1基因A10208G位点多态性与宁夏地区人群2型糖尿病存在相关性,与2型糖尿病是否合并高血压无明显关联,AGTR1基因A10208G位点多态性可能是宁夏地区人群2型糖尿病发病的遗传学易感基因. AIM: To examine whether the A10208G polymorphism in the promoter of AGTR1 gene is associated with type 2 diabetic mellitus (T2DM) and T2DM complicated with hypertension in the population of Ningxia District. METHODS: Polymerase chain reaction-restriction fragment length polymorphism (PCRRFLP) method was used to analyze the A10208G polymorphism in the promoter of AGTR1 gene in 85 cases of T2DM ( DM group), 42 cases of T2DM complicated with hypertension (DM-H group) and 224 healthy subjects (NC groups). The clinical parameters and serum markers were also measured. RESULTS: The frequencies of AA, AG and GG genotypes of AGTR1 gene promoter A10208G were significantly different between DM group and NC group ( enotype: x^2= 5.94, P 〈 0.05. Allele: x^2= 4. 03, P 〈0.05 ). The BMI and blood pressure were significantly increased in DM-H with G allele group than in DM with G allele group ( P 〈 0.01 ). CONCLUSION: The A10208G polymorphism in the promoter of the AGTR1 gene has association with T2DM in the population of Ningxia District. No association between A10208G polymorphism and T2DM complicated with hypertension is observed. It might be a genetic risk factor of T2DM in Chinese.
