Expression and significance of NF-κB and VEGF in renal cell carcinoma

摘要 目的:研究核因子κB(nuclear factor-κappaB,NF-κB)和血管内皮细胞生长因子(vascular endothelial growth facor,VEGF)在肾癌中的表达及其意义。方法:应用免疫组化方法(Envision二步法)检测40例肾癌组织及10例正常肾组织中的NF-κB P65、VEGF和CD34的表达,并与肾癌的临床分期进行比较。结果:NF-κB P65显著表达于肾癌组织中,40例肾癌组织中有26例呈阳性表达(65.0%),正常肾组织中呈阴性表达;VEGF在40例肾癌中有27例阳性表达,表达率为67.5%,显著高于正常肾组织(20.0%,P<0.05);NF-κB P65与VEGF同时表达组肾癌的微血管密度(microvessel density,MVD)明显较两者同时阴性组高(P<0.01);肾癌中NF-κB P65的表达与VEGF的表达呈正相关(P<0.01);NF-κB P65与VEGF的表达均与肾癌的临床分期有关。结论:NF-κB在肾癌中呈异常活性表达,可能通过多种途径参与肾癌的发生发展;NF-κB对于VEGF的高表达可能具有潜在的正向调节作用。 Objective: To investigate the expression and significance of nuclear factor-kappaB (NF-kB) and vascular endothelial growth factor(VEGF) in primary renal cell carcinoma(RCC). Methods: Immunohistochemistry was used to detect the expression of NF-kB p65, VEGF and CD34 in 40 cases of RCC and 10 cases of renal tissue. The correlation between the expression in RCC and clinicopathological stage was examined. Results: Twenty-six of 40 RCC were positive for NF-kB 5 protein with a 65.0% positive rate (65.0%), but there was lower expression in normal renal tissue. The positive rate of VEGF in RCC (27/ 40 ;67.5% ) was significantly higher than that in normal renal tissue( 20.0%, P 〈 0.05 ). The microvessel density(MVD) in co-expression of NF-kB p65 and VEGF group was significantly higher than that in nonexpression group(P 〈0.01 ). The expression of NF-kB p65 has a positive correlation with that of VEGF in PRCC (r=0. 498 ,P 〈0.01 ), and the expression of the two factors was correlated with clinicopathological stage. Conclusion: NF-kB was activated abnormally in PRCC. It is probably involved in the etiology of PRCC by various pathways. NF-kB may positively adjust high VEGF expression in PRCC.
