慢性非细菌性前列腺炎/慢性骨盆疼痛综合征患者外周血T细胞受体Vβ基因克隆性分析 被引量:6

摘要 目的检测慢性非细菌性前列腺炎/慢性骨盆疼痛综合征(CAP/CPPS)患者外周血T细胞受体(TCR)Vβ1、Vβ8、Vβ11和Vβ20基因表达,了解TCRVβ克隆增殖与CAP/CPPS的关系。方法用半定量逆转录聚合酶链反应(RT-PCR)检测36例CAP/CPPS患者和14例正常健康男性外周血TCRVβ1、Vβ8、Vβ11和Vβ20mRNA的表达,比较两组间的差异。结果两组TCRVβ1mRNA平均相对吸光度(A)值分别为(0.24±0.08)和(0.25±0.04),P>0.05;TCRVβ8分别为(0.26±0.07)和(0.28±0.03),P>0.05;TCRVβ11分别为(0.31±0.06)和(0.27±0.05),P>0.05;而CAP/CPPS患者外周血TCRVβ20mRNA平均相对吸光度(A)值明显高于正常对照组,分别为(0.62±0.11)和(0.29±0.04),P<0.05。结论CAP/CPPS患者外周血存在TCRVβ20优势克隆,提示自身免疫反应可能参与了CAP/CPPS的发病。 Objective To detect the expression of T cell receptor Vβ1, Vβ8, Vβ11 and Vβ20 genes in peripheral blood of chronic abaeterial prostatitis/ehronie pelvic pain syndrome(CAP/CPPS) patients in order to explore the relationship between T cell clone proliferation and CAP/CPPS. Methods Semi-quantitative RT PCR method was used to detect the expression of TCR Vβ1, Vβ8, Vβ1 and Vβ20 mRNA in peripheral blood of 36 cases of CAP/CPPS and 14 normal controls. Results There was no significant difference in the average relative absorbance(A) value of TCR Vβ1, Vβ8 and Vβ11 mRNA in peripheral blood between two groups, while the average (A) value of TCR Vβ20 mRNA was significantly higher in the CAP/CPPS group than in the normal control group E(0.62±0. 11) vs (0.29±0.04) respectively, P〈0. 051. Conclusion The preponderate clone of TCR Vβ20 was found in CAP/CPPS patients. It suggested that autoimmune reaction may contribute to CAP/CPPS.
