摘要
目的观察子宫颈癌HeLa细胞中PMS2的表达及化疗药物对其的影响。方法通过四甲基偶氮唑蓝(MTT)法及逆转录基因扩增(RT-PCR)的方法检测化疗药物顺铂(CDDP)对HeLa子宫颈癌细胞株细胞生长的影响,并选择最佳浓度观察其基因表达水平的变化。结果顺铂对于HeLa细胞的生长具有明显的抑制作用,并与药物浓度相关,尤其是在10mg/L的浓度培养24h后最为明显,细胞生长率为68.2%。在此药物浓度培养下,HeLa细胞生存能力明显下降,同时PMS2基因表达水平明显上调(P<0.01)。结论PMS2可能参与子宫颈癌的发生、发展及转移过程,对于子宫颈癌的早期诊断以及化疗药物疗效及预后的判断有一定的意义。
Objective To study the expression of PMS2 in cervical cancer cell line HeLa and the effect of chemotherapeutics. Methods The effects of CDDP on the growth of cervical cnacer cell line HeLa were observed by using MTT and RT-PCR assays. The optimal concentration of CDDP was selected to observe its effect on the expression of PMS2. Results CDDP (0.1 to 10 mg/L) could significantly inhibit the growth of HeLa cells in a dose-dependent manner. The inhibitory effects on the growth of HeLa cells cultured with 10 mg/L CDDP for 24 h were most obvious and the growth rate of the cells was 68. 2%. The viability of the HeLa cells after cultured with 10 mg/L was obviously decreased and the expression of PMS2 mRNA obviously up-regulated simultaneously (P〈0.01). Conclusion PMS2 might be involved in the development and metastasis of cervical cancer, and play some important role in the early diagnosis of cervical cancer and evaluation of the chemotherapeutic effecls and prognosis.
