摘要
目的研究源自白血病细胞系J6-1的突变型P2X7受体对白血病细胞Ramos的影响。方法用RT-PCR、共聚焦显微镜检测P2X7受体的表达、定位及BzATP刺激下受体的聚集;MTT法测定细胞增殖;流式细胞术检测细胞周期。结果突变型P2X7受体稳定表达于Ramos细胞细胞膜及细胞质;细胞增殖明显增强;300μmol/LBzATP增加细胞死亡率;100μmol/LBzATP诱导P2X7受体聚集。结论该突变体促进Ramos细胞增殖;激动剂诱发P2X7受体聚集,过量激动剂导致细胞死亡。
Objective To study the effects of mutant P2X7 receptor from J6-1 cells on the biological characteristics of leukemia cells. Methods RT-PCR and confocal microscopy were used to detect the expression, localization of P2X7 receptor and the aggregation induced by BzATP. The proliferation was measured by MTr assays. Cell cycle was studied by FACS analysis. Results The receptor can be expressed on the membrane and in the cytoplasm of Ramos cells and the expression resulted in augmented proliferation. Mortality was increased upon 300 μmol/L BzATP stimulation, whereas receptor aggregation could be observed after the stimulation with 100μmol/L BzATP. Conclusions The expression of mutant receptor can stimulate the proliferation of Ramos cells. Receptor aggregation and apoptosis could result upon the treatment with moderate and higher concentrations of BzATP.
