摘要
目的 建立用高效液相色谱法检测藏药“佐塔”中汞的含量的方法。方法 通过将“佐塔”消化后,与二乙基二硫代氨基甲酸钠(DEDTC)配合,再用高效液相色谱法对其进行分离测定。色谱条件为:Diamonsil^TM C18柱:(250mm×4.6mm,5μm),流动相:甲醇-水(90:10)(含22μmol·L^-1DEDTC),流速:1.0mL·min^-1,检测波长:272nm。结果 Hg^2+。质量浓度在21.0~210.0 μg·mL^-1,其浓度与Hg-DEDTC配合物的峰面积呈良好的线性关系,相关系数为0.9997,平均加样回收率为101.2%,RSD为2.5%。结论 本方法准确可靠,可用于“佐塔”中的汞含量的测定。
Objective To establish an HPLC method for the determination of Hg in Zuota (a Tibetan medicine). Methods A Diamonsil^TM C18 (250 mm×4.6 mm , 5 μm) column was used with a mixture of methanol-water (containing 22 μmol·L^-1 DEDTC) (90 : 10) as the mobile phase, with the speed at 1.0 mL·min^-1, and the detector at 272 nm. Results The calibration curve was linear (r=O. 999 7) within 21.0 ~ 210.0 μg· mL^-1 of Hg^2+. The recovery was 101.2% (RSD=2.5%). Conclusion The method is accurate. It is suitable for the determination of Hg in Zuota.
