摘要 目的:探讨乳腺癌组织中DNA依赖蛋白激酶(DNA dependent protein kinase,DNA-PK)的催化亚单位DNA-PKcs的表达变化与分期的相关性。方法:采用免疫组化SP法检测30例Ⅰ期乳腺癌组织,11例ⅢB、ⅢC期乳腺癌组织及19例Ⅳ期乳腺癌组织中DNA-PKcs的表达情况。结果:DNA-PKcs在Ⅰ期乳腺癌组织中和其癌旁正常组织中均呈高表达,两者表达具有一致性(P>0.05)。DNA-PKcs在ⅢB、ⅢC期和Ⅳ期乳腺癌组织中的表达明显低于Ⅰ期乳腺癌(P<0.05和P<0.01)。同时,把ⅢB、ⅢC期和Ⅳ期合并后,与Ⅰ期比较,DNA-PKcs表达差异有极显著性意义(P<0.01)。结论:DNA-PKcs的低表达在乳腺癌发生进展和远处转移中有重要意义,对乳腺癌的分期有一定指导意义,可能成为乳腺癌分期的生物学指标。 Objective: To investigate the relationship of the expression change of DNA-PKcs in breast cancer tissue with tumor staging. Methods: By using immunohistochemical SP method ,the expression of DNA-PKcs was detected in 30 cases stage Ⅰ breast cancer tissues, Ⅱ cases of stage Ⅲ B and stage Ⅲ C breast cancer tissues, and 19 cases of stage Ⅳ breast cancer tissues respectively. Results: There was no statistically significant correlation of DNA-PKcs expression levels with stage I breast cancer tissues and ttle corresponding normal tissues (P 〉 0. 05 ) , and the expression levels of DNA-PKcs in both were high. The expression levels of DNA- PKcs in stage ⅢB, stage ⅢC and stage IV breast cancer tissues were lower than those in stage Ⅰ breast cancer tissues(P 〈0.05 and P 〈 0. 01 ). The expression of DNA-PKcs between stage I and stage Ⅲ + Ⅳ was obviously different ( P 〈 0.01 ). Conclusion: The low-expression of DNA-PKcs may play an important role in the developing of breast cancer and the metastases of breast cancer. DNA- PKcs could be recognized as a biomarker for breast cancer staging system.
