摘要
目的研究人脑胶质瘤中细胞周期素E(cyclin E)的表达及其与细胞增殖及凋亡的关系。方法采用免疫组化法和末端脱氧核昔酸转移酶介导的dUTP缺口末端标记法分别检测47例脑胶质瘤中cyclin E、增殖细胞核抗原的表达和凋亡细胞,计算三者阳性单位(PU)值,统计分析三者之间及其与胶质瘤病理分级之间的关系。结果随着人脑胶质瘤病理分级的增高,cyclin E的PU值(cyclin E PU)明显升高(t=8.08,P<0.01)。增殖指数(PI)及凋亡指数(AI)亦随着脑胶质瘤恶性程度增高而升高。cyclin E PU与PI和AI呈明显正相关(r=0.895、0.703,均P<0.01)。并且,随着cyclin E的表达水平及胶质瘤病理级别的增高,PI比AI有更大的增高趋势(F=33.72,P<0.01)。结论人脑胶质瘤中cyclin E的表达与细胞增殖和凋亡、肿瘤病理学分级密切相关,很可能参与了人脑胶质瘤的发生和恶性转化。
Objective To investigate the expression of cyclin E in human brain glioma and its relationship with pathological grades, cellular proliferation and apoptosis. Methods The expression of cyclin E,PCNA and cell apoptosis were detected by immunohistochemistry and TUNEL method in 47 cases of human brain glioma and 10 specimens of normal brain tissues. The Positive Unit (PU) of cyclin E, Proliferative Index (PI) and Apoptosis Index (AI) were detected. Results With the advancing of pathological grades, the PU of cyclin E, PI and AI in human brain glioma increased significantly. (t=8.08, P〈 0.01). PI and AI were positively correlated with the PU of cyclin E (r=0.895, P 〈 0.01 and r =0.703, P〈 0.01 respectively). Moreover, with the increasing of cyclin E and the malignancy grades of human brain glioma PI had a higher raising tendency than that of AI (F =33.72,P〈 0.01 ). Conclusion Cyclin E may be involved in the formation and progression of human brain glioma.
