托瑞米芬逆转肺癌耐药细胞株SP/N多药耐药性的研究 被引量:3

摘要 目的探讨托瑞米芬(torem ifene,TOR)逆转肺癌(lung cancer,LC)耐药的机制。方法采用四氮唑盐(MTT)比色法检测托瑞米芬对肺癌耐药细胞SP/N的逆转效应及对化疗药长春瑞滨(novelbine,NVB)敏感性的影响;半定量逆转录聚合酶链反应(RT-PCR)检测TOR处理SP/N前后细胞耐药基因的表达变化;免疫组化(IHC)检测TOR对SP/N耐药蛋白表达的逆转。结果肺癌耐药细胞SP/N中可检测到mdr1 mRNA及相应蛋白P-gp的表达,无MRP和LRP表达。托瑞米芬(5μmol/L或10μmol/L)可以抑制耐药细胞SP/N的mdr1 mRNA表达水平,增加长春瑞滨对SP/N的抑制效应。结论雌激素受体拮抗药托瑞米芬可以逆转mdr1P-gp介导的肺癌多药耐药,部分恢复耐药肿瘤细胞对化疗药物(长春瑞滨)的敏感性。 Objective To study the reversal effect of toremifene(TOR)on mdrl and P-gp in SP/N and its mechanism, Methods The expressions of mulitdrug resistance genes and its proteins on SP/N treated by TOR were examined, Reversal effects of various concentrations of TOR on sensitivity to NVB in SP/N was studied by tetrazolium dye assay(MTT), Results TOR could significantly reversed mdrl gene expression in SP/N after treatment with TOR(5 μmol/L or 10 μmol/L), Conclusions TOR has some reversal effects on mdrl in SP/N and declines the expression levels of mdrl mRNA and its protein.
