摘要
目的探讨MMP-9反义寡核苷酸作为靶基因对胃癌细胞的生长抑制效应,以进一步揭示MMP-9反义寡核苷酸治疗胃癌的可能性和可行性。方法以人胃癌细胞株MGC803为研究对象,应用ASODN技术,将MMP-9ASODN转染到MGC803细胞中,利用MTT法检测不同浓度不同时间MMP-9反义寡核苷酸对MGC803细胞的杀伤活性,流式细胞仪检测分析MMP-9反义寡核苷酸对MGC803细胞周期及凋亡的影响。结果MMP-9反义寡核苷酸在体外能够抑制MGC803肿瘤细胞增殖,且这种增殖抑制作用呈剂量和时间依赖性;MMP-9反义寡核苷酸在体外能够诱导MGC803肿瘤细胞凋亡,作用与浓度和时间有关。结论MMP-9反义寡核苷酸可望成为一种人胃癌临床有效的治疗方法。
[Objective] To detect the inhibition effect of MMP-9 ASODN on gastric cells in vivo. [Methods] Gastric cancer cell MGC803 lines were cultured with MMP-9 ASODN in a wide range of concentration for various time. Cell activity were studied by MTT. The changes of cell cycle were analyzed by FCM. [Results] The proliferation of MGC803 cell could be inhibited by MMP-9 ASODN. The inhibition effect showed the time-dose relationship. Apoptosis of gastric cancer cell lines could be efficiently induced by MMP-9 ASODN in a time/dose-dependent manner. [Conclusion] MMP-9 ASODN can inhibit gastric cancer cell lines proliferation in vivo. So it may become a kind of treatment for the gastric cancer.
