摘要
目的探讨KAI1基因在人大肠癌中的表达及其与大肠癌转移的关系。方法用RT-PCR法检测45例大肠癌组织及转移灶并以正常大肠粘膜组织作为正常对照组,进行KAI1mRNA半定量分析。结果本组中40例大肠癌组织KAI1mRNA表达水平比相应的正常大肠粘膜高(P<0.01)。大肠癌组织KAI1mRNA表达水平与临床Dukes'分期呈负相关(r=-0.474,P<0.01),与有无淋巴结转移呈负相关(r=-0.566,P<0.01)。伴淋巴结转移的大肠癌KAI1mRNA表达水平比不伴淋巴结转移的低(F=20.228,P<0.01)。转移淋巴结的KAI1mRNA表达比相应的原发灶表达降低(P<0.05)。结论KAI1mRNA表达降低与大肠癌的淋巴结转移密切相关。KAI1基因在人大肠癌中起转移抑制作用。
Objective To investigate the correlation between KAI1 gene expression and metastatic behavior in human colorectal carcinoma. Methods KAIlmRNA expression was examined in 45 colorectal primary carcinomas,4 liver metastases,10 lymph node melastases by self-quantitative reverse lransciption-polymenase chain reaction, 45 normal colonic mucosa tissue samples served as control group. Results KAI1 expression was higher in primary carcinomas than in normal mucosa (40/45,P〈0.01) ,and was inversely correlated with Dukes'stage(P〈0.01) and lymph node metastasis(P〈0.01). KAI1 expression was lower in Dukes'C and Dukes' D than in Dukes' B (P〈0.05),and lower in lymph node metastases (P〈0.01). KAI1 expression was significantly different in primary colonic tumors in the presence and absence of lymph node metastasis. Conclusion The expression of KAI1 is inversely correlated with lymph node involvement, Dukes' stage. The down-regulation of KAI1 gene is associated with lymph node metastasis of colorectal carcinoma.
