原花青素对2型糖尿病大鼠局灶性脑缺血模型的保护作用 被引量:2

摘要 目的探讨原花青素(procyanidins,PC)对2型糖尿病Sprague-Dawley(SD)大鼠大脑中动脉脑缺血的保护作用及其机制。方法高脂高糖饲料喂养4w后,小剂量链脲佐菌素(Streptozotocin,STZ)一次性尾静脉注射,制备2型糖尿病大鼠。局灶性脑缺血24h后进行神经功能评分,脑组织含水量及超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果单纯缺血组与对照组比较,表现为脑含水量增加、SOD减少、MDA增加;原花青素50 mg/kg、100 mg/kg组和200 mg/kg组与单纯缺血组比较,表现为脑含水量减少、SOD增加、MDA减少。结论高血糖可进一步加重局灶性脑缺血的自由基损伤,原花青素对2型糖尿病SD大鼠局灶性脑缺血具有保护作用,其保护作用机制可能与其抗氧化和清除自由基有关。 Objective To study the protective effect and mechanism of procyanidins on focal cerebral ischemia injury of NIDDM rats. Methods By feeding fat and glucose forage for four weeks, and injecting STZ through vena caudalis in low dose, NIDDM rats were made. The neurological function was observed, and water content of brain tissue, the activity of superroxide dismutase (SOD) and the content of malondialdehyde (MDA) in brain tissue were measured after 24h focal cerebral ischemia. Results The water content of brain tissue and MDA of the focal cerebral ischemia group are higher than the control group while the content of SOD is lower. The water content of brain tissue and MDA of procyanidins groups with 50 mg/kg, 100mg/kg and 200mg/kg were lower than focal cerebral ischemia group while SOD is higher. Conclusions Hyperglycemia aggravates free radical damage of cerebral ischemi- a, and procyanidins is protective to the focal cerebral ischemia of NIDDM rats by decreasing the activity of antioxidant ability and potent scavengers of free radicals, increasing the activity of SOD, decreasing the content of MDA.
