摘要
目的探讨基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-9(MMP-9)与基质金属蛋白酶抑制因子-1(TIMP-1)在形成肝包虫囊肿周围纤维囊壁中的作用及其临床意义。方法应用免疫组织化学技术检测MMP-1、MMP-9与TIMP-1在30例肝包虫囊肿周围纤维囊壁及周围肝组织中的表达。结果MMP-1、MMP-9与TIMP-1在肝包虫囊肿周围纤维囊壁中呈现特异性分层表达,MMP-1、MMP-9与TIMP-1的阳性率在外囊中的表达率分别为6.7%、13.3%、16.7%,在靠近肝实质侧的“外膜”中MMP-1、MMP-9与TIMP-1的阳性细胞表达率分别为83.3%、70.0%、90.0%,MMP-1、MMP-9与TIMP-1在外囊和“外膜”表达的差异均有统计学意义(均P〈0.01)。结论MMP-1、MMP-9与TIMP-1主要在肝包虫“外膜”表达,外囊较少表达;MMP-1、MMP-9与TIMP-1在肝包虫外囊和“外膜”的表达,说明MMP-1、MMP-9与TIMP-1在肝包虫囊肿周围纤维化过程中起重要作用。
Objective To evaluate the role and clinical significance of the matrix metalloproteinases-1 ,-9 (MMP-1 ,-9) and tissue inhibitor of metalloproteinases-1 (TIMP-1) in the formation of the pericystic layers of hepatic hydatid cyst. Methods Immunohistochemistry was used to observed the expression of MMP-1, MMP-9 and TIMP-1 in 30 pericystic layers of hepatic hydatid cysts. Results A special delaminated expression was observed in the pericystic layer of hepatic hydatid cysts, expressions of the MMP-1, MMP-9 and TIMP-1 in the ectocyst layer near parasite 6.7 %, 16.7 %, 13.3 % were lower than those in the exo-adventitia layer near hepatic parenchyma 83.3% ,70.0%,90.0%(P〈0.01,P〈0.01).Conclusion The expressions of MMP-1,MMP-9 and TIMP-1 in exo-adventitia and ectocyst layer of hepatic hydatid cyst demonstrates the important role of MMP-1, MMP-9 and TIMP-1 in the formative process of the fibrosis near ectocyst of hepatic hydatid cyst.
