摘要
探讨树莓酮对高脂饮食喂养的单纯性肥胖大鼠的减肥作用及其作用机制。雄性SD大鼠48只随机分为4组。对照组普通饲料喂养,模型组喂高脂饲料;高、低剂量树莓酮组分别在喂饲高脂饲料4wk后予1%和2%树莓酮灌胃。8wk末处死各组大鼠,测定大鼠体重、Lee’s指数和肾周脂肪湿重;检测血脂(TC、TG、HDL-C、LDL-C)、血糖(GLU)、胰岛素(INS)、瘦素(LEP)、游离脂肪酸(FFA)和肿瘤坏死因子α(TNFα)水平,计算胰岛素敏感指数(ISI)。结果与对照组比,模型组大鼠体重、Lee’s指数和肾周脂肪质量,GLU、INS水平,血清TG、TC、LDL-C水平,血清LEP、TNFα水平明显增高,ISI,HDL-C水平,FFA水平显著降低。高、低剂量树莓酮组各项指标较模型组有明显改善。结论树莓酮可通过调解糖脂代谢紊乱、改善瘦素抵抗和胰岛素抵抗等综合作用来降低肥胖大鼠体重。
To explore the Anti-obese action of raspberry ketone and the mechanisms of action in the treatment of simple obesity in rats fed by high-fat diets. Forty-eight male Sprague Dawley rats were randomly assigned into 4 groups. The rats fed by normal food served as the controls. The rats in model group were fed with high- fat diets, and those in high and low dose raspberry ketone groups were given 1% and 2% raspberry ketone 4 wk after high-fat diets feeding respectively. The rats in every group were killed at 8 wk. Determinning rat body mass, Lee's index and perirenal fat wet weight. Blood sample were collected for the detection of blood fat(TC, TG, HDL-C, LDL-C) and blood glucose and insulinserum and leptin-and free fatty acid and tumor necrosis factor alpha, and insulin sensitivity index were calculated. Results In comparison with those of normal rats, the body mass, Lee's index and perirenal fat wet weight and the levels of Glu and Ins and the levels of TG and TC and LDL-C and the levels of LEP and TNFα were increased apparently, but insulin sensitivity index and the levels of HDL-C,free fatty acid were decreased significantly in model group. In comparison with those of model rats, the markers such as Glu, Ins, TG and TC etc. were all significantly improved in the raspberry ketone groups. Conclusion Raspberry ketone may loss weight of obesity rats by controlling glucolipid metabolic disorder and improving leptin resistance.
