摘要
用不同温度对蚕豆子叶进行短时间的非离体处理,结果表明36℃的高温最适宜子叶细胞的分裂,有丝分裂指数明显高于16~22℃(结荚期适宜温度)时的分裂指数,二者差异非常显著.用经36℃高温处理的子叶做材料进行染色体制片和分带,染色体分散好,没有细胞质干扰,带纹清晰,首次获得了蚕豆子叶染色体C-带核型图.但是,超高温(40℃)抑制细胞分裂。
Young cotyledon of Vicia faba are treated under different temperature in living plant for short time. The results show the temperature 36℃ is best suitable for the cell division of cotyledon. Under this temperature,the mitosis index is higher remarkablely than the index between 16~22℃(suitable temperature for bearing bean-pod). The results using cotyledon treated under 36℃for preparing chromosome and chromosome banding show chromosome disperse better,no coytplasm interference,and band stria is clear. The research obtain the C-bands of chromosomes karyogram of cotyledon in Vicia faba firt. Over temperature 40℃ once,the cell division would be repressed,and the chromosome structures would be destroied.
