摘要 学术背景:血管和神经有非常相似的复杂分支结构,它们常常伴行到达同一远隔靶位,这提示它们应当受到某些共同信号分子调控。有研究表明,神经轴突导向因子ephrins、slits、semaphorins及netrins调节血管内皮细胞的迁移和血管新生。目的:分析信号分子Netrins在血管形成中的作用及其在血管和神经疾病中的应用前景。检索策略:应用计算机检索Medline2000-01/2007-04与导向因子netrins与血管形成相关的文章,检索词"netrins,neuronal guidance,angiogenesis,blood vessel,endothelial cells,vasculature system,nervous system",限定文献语言种类为English。同时计算机检索万方数据库2000-01/2007-04的相关文章,检索词"血管形成,神经导向因子,Netrins,血管,内皮细胞,神经系统,血管系统",限定文献语言种类为中文。初检索到相关文献381条,纳入标准:有关神经导向因子Netrins在血管形成中作用的文章,筛除重复性研究和综述文献,最后共纳入文章30篇,其中有关血管形成的文章9篇,netrins在血管形成过程中作用的21篇。文献评价:所有文献均为RCT发表,均是来源于动物实验。资料综合:内皮祖细胞存在于各种成年动物和人的骨髓及外周血中,也可由骨髓、成人外周血或脐血中的单核细胞在体外培养、扩增得到。内皮祖细胞参与血管新生和缺血损伤后的血管修复,在这一过程中,需要生长因子和信号分子参与。其中神经导向因子Netrins不仅参与神经的生长还参与血管新生,Netrin-1对人动脉和微脉管系统初期不同内皮细胞的促有丝分裂前、粘附前和迁移前有积极效应。Netrins能恢复神经和血管功能,Netrin-1、Netrin-4与血管内皮生长因子都有促血管形成作用,但以Netrin-1在恢复神经传导速度方面更具有优越性,可能是因为具有内皮和神经的双重生物学效应。结论:Netrins具有促进神经和血管的生长及再生能力,这为许多退行性疾病的治疗提供了广阔的前景,尤其是对糖尿病引起的血管和神经并发症的治疗具有重要的临床意义。 BACKGROUND: Nerves and blood vessels resemble each other in their ability to form branching networks and accompany to get to their target. It is indicated that they will be regulated by some common signaling molecules. Studies have shown that ephrins, slits, semaphorin and netrin can regulate the migration of vascular endothelial cells (VEC) and the angiogenesis. OBJECTIVE: To analyze the effect of Netrins in angiogenesis and its prospect application in nerves and blood vessels diseases. RETRIEVAL STRATEGY: The Medline Database was undertaken to identify the relevant articles on the neuronal guidance factor netrins and developmental angiogenesis published from January 2000 to April 2007 with the key words of "netrins, neuronal guidance, angiogenesis, blood vessel, endothelial cells, vasculature system, nervous system" in English. Simultaneously, relevant articles were retrieved in Wanfang Database published from January 2000 to April 2007 with the same key words in Chinese. 381 literatures were checked primarily. Inclusive criteria included relevant articles on the role of Netrins in angiogenesis. Exclusive criteria included repetitive research and review article. A total of 30 articles met the inclusive criteria. Of them, 9 articles were on process of angiogenesis and 21 articles were on the role of netrin in angiogenesis. LITERATURE EVALUATION: All articles on animal tests are RCT. DATA SYNTHESIS: Endothelial progenitor cells lie in peripheral blood and bone marrow in all kinds of adult animals and human. It may be harvested from cultured mononuclear cells. Endothelial progenitor cells participate in blood vessel angiogenesis and injured vascular repair. This process needs growth factors and signaling molecules. Nerve guidance Netrins play major roles in neuroal growth and angiogenesis. Netrin-I plays positive effect in the vascular formation process from endothelial cells of human artery and microvessel before karyomitosis, adherence and migration. Netrins can recovery the function of neuron and vascular, such as Netrin-l, Netrin-4. But netrin-1 has superiority in the nerve conduction velocity recovery. Perhaps it has dual biological effects of endothelium and nerve. CONCLUSION: Netrins promotes the growth and regeneration of nerve and vessel, which have an important clinical significance and widest applicable prospect in many degenerative diseases, such as diabetes mellitus complications of nerve and blood vessel.ured vascular re.
