磷酰化壳聚糖仿生衍生物的合成及其与DNA的相互作用 被引量:2

摘要 采用Antherton-Todd反应制备了两种水溶性的磷酰化壳聚糖仿生衍生物:两性的N-磷酸胆碱磷酰化壳聚糖(N-PCCs)和带正电荷的N-乙基磷酸胆碱磷酰化壳聚糖(N-PC+Cs),并用紫外吸收光谱(UV)和圆二色光谱(CD)研究了两种胆碱磷酰化壳聚糖衍生物与小牛胸腺DNA的相互作用.结果表明:壳聚糖衍生物的电荷状态对其与DNA的相互作用影响很大.在pH=7.2的中性条件下,两性的磷酸胆碱壳聚糖衍生物与DNA作用不明显,而带正电荷的乙基磷酸胆碱磷酰化壳聚糖衍生物与DNA有强烈的静电作用. Two kinds of water-soluble biomimetic chitosan derivatives with a phosphoramide linkage between glucosamine and choline were prepared through Atherton-Todd reaction under the mild conditions, including zwitterionic phosphorylcholine ( PC ) bound chitosan ( N-PCCs ) and acetyl-phosphorylcholine bound chitosan ( N-PC + Cs ) with positive charge. The interaction between chitosan derivafives and calf thymus DNA were inversigated by UV absorption spectra and circular dichroism ( CD ) spectra. The results indicated that the interaction between chitosan derivatives and DNA was related to the charge state of chitosan derivatives. In the neutral solution ( pH = 7.2 ), no obvious interaction between N-PCCs and DNA was observed, while there's strong static electric interction between N-PC + Cs and DNA.
