摘要 采用高盐低pH缓冲液进行粗提,粗提液经阳离子交换层析-RP HPLC-Sephadex G25凝胶过滤-C18RP-HPLC纯化,得到纯化多肽。尿素-Tricine-SDS PAGE测定其分子量为3.8kDa,215nm具最大吸收。对Ecoli、S.aureus、B.proteus和B.subtilis有强的抗菌活性。 To isolate and purify β-defensin(SBD) from the small intestine of Mongolia Ovis aries, acid soluble extract of Mongolia Ovis aries small intestine was obtained. Then it was further purified by cation exchange chromatography, RP- HPLC, G-25 gel filtration, and RP-HPLC. The antibacterial activities of the extracts from every step were analyzed by agarose radial diffusion assay. The molecular weight of the purified SBD was determined to be 3.8 kDa by Tricine- SDS-PAGE. SBD-2 from Mongolia Ovis aries small intestine was able to effectively kill E. coli, S. aureus, B. subtilis and Bacillus proteus. SBD-2 has an important part in innate immunity in Mongolia Ovis aries.
