Expression of STAT3 mRNA in the hippocampus following transient global cerebral ischemia-reperfusion

摘要 目的研究鼠海马组织中信号转导与转录激活子-3(STAT3)mRNA在短暂性全脑缺血-再灌注过程中的表达变化。方法将健康雄性SD大鼠25只,随机均分为全脑缺血-再灌注4、24、48、72h组和假手术组。以"双侧颈总动脉阻断加全身低血压法"建立大鼠短暂性全脑缺血模型,采用实时荧光定量PCR技术观察大鼠缺血性脑损伤海马组织中STAT3 mRNA的变化。结果短暂性全脑缺血后海马组织STAT3 mRNA的表达增强,再灌注24h达高峰。结论短暂性全脑缺血-再灌注后STAT3 mRNA的活化及超量表达可能介导了缺血神经细胞信号传导过程,并参与了脑神经细胞损伤的病理生理过程。 Objective To observe the expression and dynamic changes of signal transducers and activators of transcription3 (STAT3)mRNA after transient global ischemia-reperfusion in the hippocampal CAI region. Methods Transient global ischemia was induced in rats by bilateral common carotid artery(CCA) occlusion combined with hypotension. Animals were allocated to groups of sham (group E),4 h(group A), 24 h(group B),48 h(group C) and 72 h (group D)after transient global cerebral ischemia-reperfusion. The expression of STAT3 mRNA was detected with RT-PCR. Results Compared with the sham operation group, the expression of STAT3 mRNA increased significantly at 4 h,arrived peak at 24 h,and tended to decrease at 72 h after reperfusion. Conclusion The enhanced gene transcription of STAT3 mRNA might indicate the transmission course in ischemic nerve cell signal,and plays a role in the pathophysiology in ischemia cerebral injury.
