摘要
目的探讨血红素加氧酶-1(HO-1)对大鼠原位肝移植后BCL-2和VCAM-1的影响。方法建立大鼠原位肝移植排斥模型,将大鼠分为三组:对照组(A组),ZnPP组(B组)和CoPP组(C组)。三组分别于3d和7d处死,取肝脏标本。荧光实时RT—PCR检测HO-1及VCAM-1的表达,免疫组化方法(SP)检测BCL-2的表达。结果(1)HO-1在C组表达明显升高,B组不明显,A组介于两组之间。(2)VCAM-1在3d时C组与A或B组相比有明显统计学意义(P〈0.05),A组与B组相比无统计学意义(P〉0.05)。7d时B组与C组相比有明显统计学意义(P〈0.05),A组与B或c组相比无统计学意义(P〉0.05)。(3)BCL-2免疫组化染色阳性面积百分率(x^-±s,%)统计学分析显示,3d和7d时C组与A或B组相比较有明显的统计学意义(P〈0.05),A组和B组相比无统计学意义(P〉0.05)。结论HO-1过表达上调BCL-2的表达,抑制VCAM-1的表达。
Objective To investigate the effect of HO-1 on BCL-2 and VCAM-1 during orthotopic liver transplantation (OLT) in rats. Methods After the model of OLT with acute rejection was established in rats, the rats were divided into the control group (group A), group ZnPP (group B) and group CoPP (group C). On the 3rd and 7th d after the transplantation, the recipient rats were killed and liver samples collected to determine the expression of BCL-2 with immunohistochemistry and that of HO-1 and VCAM-1 with fluorescent real-time RT-PCR. Results (1)The expression level of HO-1 was from high to low in order from group C, group A to group B. (2) As for VCAM-1 expression level on the 3rd after operation, it was significantly different between group C and group A or group C and group B (P〈0.05). There was no marked difference between group A and group B (P〉 0.05). On the 7th d after the operation, its expression level was significantly different between group B and group C (P〈0.05) but not different between group A and group B or group C (P〉0. 05). (3) On the 3rd and 7th d after operation, the expression area percentage of BCL-2 was markedly different between group C and group A or group B (P%0.05). There was no remarkable difference between group A and group B (P〉0.05). Conclusion The expression of HL-1 in rats with acute rejection after OLT can up-regulate the expression of BCL-2 and down-regulate that of VCAM-1.
