摘要
目的探讨缺氧环境对人肝癌细胞侵袭能力的影响。方法用Transwell小室筛选高侵袭能力的HepG2细胞株,用氯化钴(150μg/ml)对该高侵袭细胞株进行化学诱导缺氧培养,分别于培养24,48,72h后,用Western印迹法检测HIF-1α蛋白的表达,用RT—PCR方法检测CD44v6 mRNA的相对表达量,统计学分析两者的改变。结果随着缺氧培养时间的增加,HIF-1α蛋白和CD44v6 mRNA的表达量的改变具有显著性差异,并且两者呈同步改变。结论缺氧可以通过增加肝癌细胞的迁徙能力促进肿瘤的转移。
Objective To explore the effect of hypoxia on the ability of invasion of HepG2 cell. Methods Using Transwell microchemotaxis chamber system, we separated H1 cell line which has the ability of higher invasion and cultured the H1 cell llne under hypoxic condition produced by cobalt chloride. The expression of the protein of HIF-1α and CD44v6 mRNA was determined by Western blotting and RT-PCR, respectively, after culture for 24 h, 48 h, 72 h. Results After culture under hypoxia, the expression of HIF-1α protein and CD44v6 mRNA was stepped up correlating to the time of culturing, and the changes were synchronous correlating to the degree of hypoxia. Conclusions The upregulation of CD44v6 mRNA can be induced by hypoxia. The protein of HIF-1α may be critical to the upregulation of the expression of the CD44v6 mRNA and play an important role in the neoplasm invasion by metastatic behavior not only stimulating the expression of VEGF.
