摘要
目的观察复方黄黛片含药兔血清对NB4细胞株的影响。方法以兔为实验血清的供体,以给药与否为分组依据,采用双道原子荧光法检测血清中砷剂的浓度,以细胞抑制率及调亡率为观察指标,对实验结果进行分析。结果(1)给药前、后兔血清中砷剂差异显著;(2)两组兔血清对NB4细胞株生长均有抑制作用,且对NB4细胞株生长的抑制作用具有一定的浓度及时间依赖性;(3)两组兔血清均可诱导NB4细胞株发生凋亡,且该作用具有一定的浓度及时间依赖性。结论(1)复方黄黛片含药兔血清可明显抑制人急性早幼粒细胞白血病NB4细胞株的生长,抑制作用呈一定的浓度、时间依赖性;(2)复方黄黛片含药兔血清可诱导人急性早幼粒细胞白血病NB4细胞株发生凋亡,该作用呈一定的浓度、时间依赖性;(3)中药血清药理学方法可用于深入研究复方黄黛片抗白血病的作用机理。
Objective: To explore the effects of leporid blood serum with compound realgar natural indigo tablet on cell line NB4. Methods: The experimental rabbits were took as the provider of the animal blood serum. The blood serum was divided into two groups based on whether the rabbits were given Compound Realgar Natural Indigo Tablet (CRNIT). AFS - 230a double path atom fluorescence photometer was used to detect the concentration of arsenic in the rabbits blood serum. The inhibition rations and apoptosis ratios were the observational index. Results: (1) The concentration of arsenic in the rabbits blood serum with and without the medicine had significant difference. (2) The two groups of serum all had the inhibitory effect on the growth of NB4 cells which depended on the medicine concentration and effect time. (3) The two groups of serum all can induce the apoptosis of NB4 with positive relations of the concentration and effect time. Conclusion: (1) The rabbits blood serum with CRNIT can obviously restrain the growth of NB4 cells and the inhibitory effect depends on the concentration and effect time. (2) The rabbits blood serum with CRNIT can induce the apoptosis of NB4 cell line and the apoptosis ratios depend on the concentration and effect time. (3) The method of Chinese medicine serum pharmacology can relatively reflect the effect of medicine in the body, can indicate the collective function of the four components in CRNIT in the round and it is feasible to lucubrate the mechanism of the effect of CRNIT on leukaemia.
