摘要
以AdEasy腺病毒载体系统为基础,建立了一套简单有效构建编码结核分枝杆菌抗原的重组腺病毒载体的方法.依此方法挑取的重组腺病毒质粒阳性率高达90%,线性化的重组腺病毒质粒转染293A细胞24 h后可观察到报告基因GFP的表达.
On the basis of AdEasy Adenoviral System, a simple and effective method for constructing the recombinant adenoviral vector encoding the antigen of M. tuberculosis was established. Up to 90% of the picked crones were positive. The green fluorescence from GFP (green fluorescence protein) reporter of 293A cells transfected with the linearized recombinant adenoviral plasmid was observed after 24 h.
