炙甘草汤对大鼠在体心肌缺血-再灌注损伤后左心功能及抗氧化酶的影响 被引量:41

The Effects of Zhi Gan Cao Decoction on the Left Cardiac Function and Antioxidative Enzymes in Ischemia-Reperfusion Injured Rats

摘要 目的探讨炙甘草汤对大鼠在体心肌缺血再灌注损伤后左心功能及抗氧化酶的影响。方法大鼠随机分为两组,生理盐水对照组和炙甘草汤处理组,分别用生理盐水和炙甘草汤灌胃4 d后,造成缺血-再灌注模型,观察再灌注10,20min后左心室内压力峰值(LVSP)、左心室舒张末期压(LVEDP)和左心室内压最大变化速率(±dp/dtmax);血中超氧化物歧化酶(SOD)及过氧化氢酶(CAT)活性、活性氧(ROS)及丙二醛(MDA)含量的变化。结果再灌注10 min时,炙甘草汤处理组与生理盐水对照组比较,LVSP及±dp/dtmax明显升高(P<0 01,P<0 05),LVEDP明显降低(P<0 01);再灌注20min后,炙甘草汤处理组与生理盐水对照组比较,LVSP明显升高(P<0.01)、±dp/dtmax变化不显著,LVEDP降低(P<0 05);炙甘草汤能明显提高再灌注10 min时血中SOD活性(P<0.01),降低MDA和ROS的含量(P<0.01)。结论炙甘草汤能明显提高缺血-再灌注损伤后的左心功能,可提高血中SOD活性,降低MDA和ROS的含量。 Objective To investigate the effects of Zhi Gan Cao Decoction on left cardiac function and antioxidative enzymes in ischemia - reperfusion injured rats. Methods Rats were divided into two groups:control group and treating group, After the rats were adminstrated with 0.9% NaCl or Zhi Gan Cao Decoction for 4 days, the myocardial ischemia - reperfusion injury model was established. Left ventricular systolic pressure(LVPS) , left ventricular end- diastolic pressure(LVEDP) and ± dp/dtmax after reperfusion 10min, 20min were measured; the activities of SOD and CAT, the levels of ROS and MDA after reperfusion 10min. Results Zhi Gan Cao Decoction could obviously increase LVSP, ±dp/dtmax after reperfusion 10min(P 〈0.01 ,P 〈0.05), decrease LVEDP ( P 〈 0.01 ) after reperfusion 10rain ; increase LVSP after reperfusion 20rain( P 〈 0.01 ), decrease LVEDP ( P 〈 0.05 ) after reperfusion 20rain, but had ±dp/dtmax ; Zhi Gan Cao Decoction could obviously increase the activity of SOD, decrease the levels of ROS and MDA( P 〈 0.01 ). Conclusion Zhi Gan Cao Decoction can obviously improve the left cardiac function in ischemia- reperfusion injured rats, increase the activity of SOD and decrease the levels of ROS and MDA.
