摘要
在热变性法提取猪血中SOD工艺中,比较了Cu2+和Zn2+对SOD的保护作用和沉淀杂蛋白的作用,试验表明:这两种离子的硫酸盐对SOD的保护效应要优于氯盐;当Cu2+和Zn2+浓度分别为2mmol/L和3mmol/L共同作用时对SOD的保护效果最好,SOD比活力比空白对照组的提高了78.4%,杂蛋白沉淀量也提高了21.2%;而在这个Cu2+和Zn2+浓度下,热变性温度65℃时,SOD的比活力最高,杂蛋白的沉淀效果也较好。
Crude swine superoxide dismutase was treated by heating denaturalization with different concentration of Cu^2+ and Zn^2+ and under various temperatures for finding the best condition of purifying swine superoxide dismutase. It was found that sulfates of Cu^2+ and Zn^2+ are better than chlorates and 2 mmol/L Cu^2+ and 3 mmol/L Zn^2+ is the best for protecting SOD. Under this condition, the relative activity of SOD is enhanced by 78.4% over and that of the control and the precipitation of miscellaneous proteins is also improved by 21.2%. At this concentration of Cu^2+ and Zn^2+ , the best heating temperature is 65 ℃.
