摘要
采用磷脂酶A1水解蛋黄卵磷脂,通过单因素实验和正交实验确定该酶水解蛋黄卵磷脂经济合理的工艺条件为:反应温度50℃,反应时间5 h,加酶量0.40 IU/g,起始pH 6.0,底物质量浓度80 g/L;利用电喷雾飞行时间质谱仪分析水解前后蛋黄卵磷脂组分,磷脂酶A1水解Sn-1位脂肪酸,生成含Sn-2位不饱和脂肪酸的溶血磷脂,反应过程中有酰基位移发生。
The hydrolysis of egg yolk lecithin in aqueous phase was studied. The economical and reasonable conditions of the reaction, investigated by the single factor experiment and orthogonal experiment, were as follows: reaction temperature 50 ℃, reaction time 5 h, phospholipase A1 dosage 0. 40 IU/g, initial pH 6. 0, mass concentration of substrate 80 g/L . The compositions of lecithin and hydrolyzed lecithin were analyzed by Electrospray Ionization - Time of Flight - Mass Spectrometry ( ESI - TOF - MS) , and the results showed that the phospholipase A1 hydrolyzed Sn - 1 position fatty acids to obtain lysophospholipids containing Sn -2 position unsaturated fatty acids. Acyl migration was observed during the reaction.
