摘要
目的探讨肝细胞生长因子(HGF)对大鼠心脏移植后细胞凋亡的影响。方法建立大鼠异位心脏移植模型,将32只受体SD大鼠随机分为两组:对照组术后注射生理盐水1 ml.kg-1.d-1;实验组术后15 d注射HGF50μg.kg-1.d-1。用HE染色和TUNEL技术检测移植心脏切片进行排斥反应的病理分级,计算凋亡指数,并观察移植存活时间。结果与对照组比较,HGF能明显减少心肌细胞凋亡(P<0.05),减轻移植物的组织损伤,延长移植物存活时间(P<0.05)。结论外源性HGF能抑制移植心脏细胞凋亡,发挥抗排斥反应作用,显著延长移植物存活时间。
Objective To study the effect of hepatocyte growth factor on cardiomyocyte apoptosis during acute heart transplantation rejection in rat. Methods Heterotopic heart transplantation model in rats was set up. Thirty-two heart transplantation SD rats were divided into two groups randomly:control group ( normal saline 1 ml/kg per day) ,HGF group ( HGF 50 μg/kg per day during the initial 15 days after transplantation). Histopathological rejection grade and apoptotic index were analyzed. Results Compared with the control group, myocyte apoptosis reduce markedly (P 〈 0.05 ), the damage to the graft tissue was reduced, and allografted heart survival significantly prolonged in HGF-treated group (P 〈 0.05). Conclusion Exogenous HGF can inhibit cardiomyocyte apoptosis apparently and suppress cardiac allograft rejection. It could prolong the survival time of transplanted hearts.
