摘要
目的寻找腹主动脉瘤(AAA)患者血清中有助于疾病诊断及病程监控的标记性蛋白质。方法利用双向电泳(2-DE)及基质辅助激光解吸/电离飞行时间质谱(MALDI—ToF-MS)技术对腹主动脉瘤患者及年龄性别匹配之健康对照的血清样本进行蛋白质组的比较分析。以Western blot验证目标蛋白在患者及对照血清中的表达差异。结果发现由一簇6个蛋白质点组成的特异性的蛋白质2-D谱图出现于对照组的全部五个血清样品中,而在病例组血清中处于低表达或检测不出。质谱鉴定这一簇蛋白质点大部分为触珠蛋白(Hp)及其前体(Hpr),Western blot验证了Hp在腹主动脉瘤患者血清中的低表达。结论触珠蛋白的低表达有助于粥样硬化性腹主动脉瘤的诊断和病程监控。
Objective To screening out novel serum biomarkers associated with abdominal aortic aneurysm (AAA) to facilitate ealy diagnosis and disease management. Methods Serological comparison analysis was performed between AAA patients and age and sex matched healthy controls, using two-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry ( MALDI-ToF-MS). Western blot was performed to confirm the differential expression of target protein between patients and controls. Results A 2-D pattern composed of 6 protein spots was found in samples from all 5 controls, most of which were identified as haptoglobin (HP)/haptoglobin related protein precursor (Hpr). They were weakly expression or completely lost in samples from AAA patients. Western blot confirmed the decreased expression of haptoglobin in sera from AAA patients. Conclusion Lower expression of serum haptoglobin may contribute to the progression of AAA and may serve as a diagnostic marker of the disease.
