摘要
目的探讨重组人肝细胞生长因子(rh-HGF)对大鼠心脏移植冠状血管内皮细胞免疫原性的影响。方法近交系健康雄性Wistar大鼠80只和雄性SD大鼠40只,建立腹腔异位心脏移植模型。实验分3组:同系移植组,异系移植包括对照组(CsA组)和实验组(HGF组)。分别于术后15、60d采集各组标本,沿冠状动脉切取心肌组织,进行免疫组织化学检测冠状血管内膜细胞间黏附分子(ICAM)-1的表达,逆转录-聚合酶链反应(RT-PCR)检测冠状血管内膜γ-干扰素(IFN-γ)mRNA表达,以及观测冠状血管病理变化。结果实验组移植心脏冠状动脉内皮细胞完整,管腔轻度狭窄,少量淋巴细胞和单核巨噬细胞浸润,病理改变明显轻于对照组。免疫组织化学测定实验组ICAM-1(0.86±0.06)、(1.26±0.19)明显低于对照组(1.39±0.20)、(2.05±0.31)表达(P〈0.01);RT-PCR测定实验组IFN-γ mRNA(1.38±0.22)、(1.06±0.12)明显低于对照组(1.97±0.34)、(1.63±0.25)表达(P〈0.01)。结论rh-HGF可能具有免疫调节功能,抑制移植心脏冠状血管内皮细胞免疫活性,减轻内皮细胞损伤,降低CAV发生易感性。
Objective To investigate the effects of human recombinant hepatocyte growth factor (rh-HGF) on immunomodulation of allograft vascular endothelial cells after cardiac transplantation in rats. Methods Heterotopic heart transplantation model was established in abdominal cavity of rats. The recipient rats were divided into three groups : isograft group ( n = 20) , control group ( CsA group, allograft n = 20) ,and experimental group (HGF group,allograft n =20). The cardiac grafts were harvested at 15th and 60th day after transplantation. The cross-sections of vascular tissues were used for immunohistochemistrical staining of ICAM-1, and the expression of IFN-γ mRNA in intima of allograft vessels was detected by reverse transcription-polymerase chain reaction ( RT-PCR). The pathologic changes of allograft coronary vessels were observed by histopathological method. Results The allograft coronary artries showed minimally intimal thickening, the endothelium and internal elastic lamina remained almost intact, and lymphocyte and mononuclear cells diffuse infiltration reduced in experimental group after transplantation. The expression of ICAM-1 was significantly downregulated in intima of allograft vessels after transplantation in experimental group ( 0.86 ± 0.06,1.26 ± 0.19) as compared with that in control group ( 1.39 ± 0.20,2.05 ± 0.31 ) ( P 〈 0.01 ). The expression of IFN-γ mRNA was significantly decreasded in experimental group ( 1.38 ± 0.22,1.06 ± 0. 12) as compared with that in control group ( 1.97 ± 0.34,1.63 ± 0.25 ) ( P 〈 0.01 ). Conclusion rh-HGF may play an immunomodulative role, suppress the activation of endothelial cells and decrease susceptibility to the development of CAV.
