摘要
目的采用高通量基因芯片,检测胃癌组织基因表达的变化,结合流行病学发病因素调查和临床病理等资料,分析胃癌异常基因表达与发病易感因素和临床病理的相关性。方法收集23例胃癌患者病变及配对非病变胃黏膜组织,用含有13824个基因或表达序列标签(EST)序列的SBC—Human2.3基因芯片进行基因表达谱检测;调查胃癌相关流行病学易感因素.结合内镜检查和外科手术病理等临床资料进行统计学分析。结果筛选出符合条件的基因53个,其中有12个目前功能不明的基因/EST序列。部分基因异常表达与少食新鲜蔬菜、水果和生活压力过大等易感因素相关(P〈0.05),且与Borrmann分型、幽门螺杆菌感染和淋巴结转移数目多(≥7个)等因素相关(P〈0.05)。结论胃癌基因表达既有一定相似性,又有异质性,其表达与易感因素及肿瘤生物学行为特点相关。分析胃癌基因表达谱有助于追踪癌前状态的转归、发展,了解胃癌的发病与生物学特征。
Objective To investigate the gene expression profile of gastric cancer associated with epidemiologic predisposing factors, endoscopic findings and pathological features. Methods Paired specimens from 23 patients with gastric cancer were studied with gene chips containing 13,824 genes/ESTs. The epidemiological data, endoscopic and pathologic findings were collected. Results Fifty-three specific genes were selected. Among those, 12 new genes/ESTs functions were unknown. The differential expressions of these genes were associated with fresh vegetables and fruits intake, the Borrmann's classification, H. pylori infection and the metastasis of lymph nodes( ≥ 7). Conclusions Gene expression of gastric cancer is correlated with epidemiological factors and biologic behaviors. The analysis of gene expression profile is helpful to elucidate the underlying mechanism of gastric cancer, its development and outcome.
