Expression of apoptosis-associated genes in hypertropic scar fibroblast by substance P and its receptor antagonist

摘要 目的 探讨感觉神经肽(substance P,SP)及其受体拮抗剂对增生性瘢痕(hypertrophic scar,HS)中成纤维细胞(fibroblast,FB)凋亡相关基因表达的影响。方法 体外培养HS及正常皮肤(normal skin,NS)的FB,免疫组化检测在SP及SP受体拮抗剂作用下FB的增殖细胞核抗原(proliferating cell nuclear,PCNA)、bax、bcl-2的表达变化。结果 SP可以促进FB的PCNA、bcl-2表达,抑制bax表达,在HS中SP作用强于NS,且主要由NK1受体介导起作用。结论SP参与FB凋亡相关基因的表达。 Objective To investigate the expression of apoptosis-associated genes in hypertropic scar fibroblast by substance P and substance P receptor antagonist. Methods The fibroblasts deriving from hypertropic scar and normal skin were cultured in media containing SP and SP NK1/ NK2 receptor antagonist. Then the expression of PCNA, bax and bcl-2 in fibroblasts was detected by immunohistochemistry. Results SP enhanced the expression of PCNA and bcl-2, and inhibited the expression of bax. The effect of SP on hypertropic scar was more significantly than normal skin. The major functional receptor was NK1 to substance P. Conclusion The substance P can affect the expression of apoptosis-associated genes in hypertropic scar.
