摘要
本文利用正交实验的方法,选取乙腈浓度、TFA浓度、洗脱速度三个因素,对高效液相色谱法分析降血压肽AHP的工艺条件进行了研究。色谱图显示,最佳分离条件为ACN浓度12%(v/v),TFA浓度0.03%(v/v),洗脱速度0.8 mL/min,保留时间为10.30min;或ACN浓度12%(v/v),TFA浓度0.05%(v/v),洗脱速度1.0 mL/min,保留时间为11.41 min。高效液相色谱分析条件的优化,有利于AHP的检测和分析,从而帮助分离纯化研究的顺利进行。
Application of RP-HPLC in analysis of antihypertensive peptides was optimized through orthogonal experiment. The optimal conditions for analysis were as follows: 12% (v/v) of ACN, 0.05% (v/v) of TFA, elution rate 1.0mL/min; or 12% (v/v) of ACN, 0.03% (v/v) of TFA, elution rate 0.8mL/min. The AHP could be separated completely from other materials under these two conditions. This result is helpful in analysis of antihypertensive peptides, in order to make sure the study in isolation and purification.
