摘要
本试验主要对一株产木聚糖酶B45菌株的液体发酵培养基组分进行优化。通过单因素和正交试验确定了所试因素的最佳组合,并对其酶学性质进行研究。结果表明:B45菌株液体发酵培养基中的最优组分为:碳源(玉米芯)2%,氮源(玉米粉)0.2%,无机盐(KH2PO4+MgSO4)0.1%,微量元素(FeSO4)0.0015%。木聚糖酶的最适反应pH为5.0,最适反应温度为50℃,有较好的热稳定性,但不耐强酸(pH≥4)和碱性(pH≤7)。
In this article,the liquid fermentation medium components of xylanase producing B45 strain was optimized. The results of single-factor and orthogonal experiment showed that the optimum medium were corncob 2 %,corn-powder 0.2 %, (KH2PO4+MgSO4)0.1 % ,FeSO4 0.0015 %.The optimal reaction conditions of xylanase were in pH 5.0 and under 50℃ with high thermal stability.
