摘要
目的:探讨宫颈癌组织放射治疗前后Aurora-A、p21基因的转录表达及其变化与临床放射治疗之间的关系。方法:运用RT-PCR的方法检测了103例宫颈癌组织中Aurora-A、p21的表达情况,放射治疗前后的变化。结果:103例宫颈癌组织中89例组织中(86.40%)检测至Aurora-A表达。经放疗后,63例(61.17%)表达减弱、7例(6.80%)表达增强,19例(18.45%)无明显变化,14例(13.59%)放疗前没有检测到Aurora-A表达的宫颈癌组织仍然没有检测到其表达。有75例(72.82%)组织表达p21。放射治疗后58例((56.31%)表达减弱,3例(2.91%)没有明显改变,14例(13.59%)表达增强。其余28例宫颈癌组织(27.18%)放疗前后均没有检测到p21表达。结论:放射治疗可以引起宫颈癌组织中Aurora-A和p21基因表达呈现降低趋势。因此,我们推测Aurora-A可能通过调节细胞周期及相关蛋白p21和c-myc等的功能,而与放射治疗效果存在一定联系。
OBJECTIVE: To investigate the expressions of Aurora - A and p21 genes in cervical cancer patients before and after receiving radiotherapy and the correlation between gene expression change and radiotherapy. METHODS: RT - PCR was applied to detect the expressions of Aurora A and p21 in cervical cancer tissues of 103 cases before and after radiotherapy. RESULTS: Of the 103 cases, 89 (86.40%) had expressions of Aurora - A. After radiotherapy, the expression of Aurora - A was down - regulated in 63 (61.17%), up - regulated in 7 (6.80%), and it was of no significant change in 19 ( 18.45 % ) ; however, there was still no expression of Aurora - A in the 14 cases ( 13.59 % ) whose expression of Aurora-A before radiotherapy was not detected. Of the total 103 cases, 75 (75.82%) had expression of P21. After radiotherapy, the expression of p21 was down - regulated in 58 (56.31% ), up - regulated in 14 ( 13.59% ), and it was of no significant change in 3 (2.91%), however, there was still no expression of p21 in the 28 cases (27.18%) whose expression of p21 before radiotherapy was not detected. CONCLUSION: Down - regulation of the expressions of Aurora - A and p21 in cervical cancer patients after radiotherapy suggests that the correlation of Aurora - A with radiotherapeutic efficacy is possibly achieved through regulating the cellular cycle and the functions of p21 and c - myc etc.
