摘要
以硝本地平片(Ⅱ)为对照,研究了硝苯地平贴片(Ⅰ)对自发性高血压大鼠(SHR)的降压作用。结果表明.Ⅰ(40mg/kg、20mg/kg、10mg/kg,皮肤给药)能显著降低SHR的收缩压(SBP)、舒张压(DBP)和平均动脉压(MAP),作用强度与剂量呈正相关.高剂量(40mg/kg)皮肤给药60min后,SBPDBP、MAP分别降低14.80%、16.64%和15.99%,至240min时,SBP、DBP、MAP分别降低了32.47%、41.29%和35.02%.中剂量(20mg/kg)、低剂量(10mg/kg)皮肤给药后亦有显著的降压作用,但降压幅度低于高剂量组。贴片对心率(HR)的影响比片剂组小,与给药前相比,贴片组动物HR在给药120min后有显著降低(P<0.05,P<0.01)。Ⅰ作用强度比Ⅱ(10mg/kg,十二指肠给药)弱,但作用较为平稳。Ⅰ和Ⅱ用药后动物心电除P-P间期有不同程度延长外,无其它明显变化。
Effects of nifedipine adhesive patch (1) and nifedipie. tablet(Ⅱ) on blood pressure and heart rate were studied in,pontaneously hypertensive rats (SHR). The results showed that administration of I (40, 21, 10mg/kg, by derma) caused a significynt decrease in systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean arteria pressure (MAP) with good dose-dependent relationship. At 60 min and 240 min after administration of I (40 mg/kg),SBP, DBP and MAP decreased about 14.80K, 16.64W, 15.99W and 32.47%, 41.29%,35.02% respectively. Thc untihypertensive effects of I (20, 10 mg/kg) were weaker than that of I (40 mg/kg), but were also obvious. The hypotensive cffects of I were weaker and steadier than that of Ⅱ (10 mg/kg, by duodenum). In addition,I exerted smaller influenca upon HR than Ⅱ did. No markedly changes except for longer P-P intorval in electrocardiograph were found after either I or Ⅱ was given.
