摘要
目的观察杏多糖与阿魏蘑菇醇提物对食管癌细胞Eca9706的凋亡作用及其作用机制。方法采用体外细胞培养,以免疫细胞化学和分子生物学技术观察10-1g/ml杏多糖与阿魏蘑菇醇提物对食管癌细胞细胞凋亡及Bcl-2、Bax表达的影响。结果免疫细胞化学结果显示,食管癌细胞经两种受试物处理24h后,Bax的表达较处理前有所增加;Bcl-2表达未见下降,反而有增高的趋势。经HE染色,在光学显微镜下可见,经10-1g/ml的杏多糖与阿魏蘑菇醇提物干预后的食管癌细胞出现了凋亡细胞的形态学特征;TUNEL测试结果显示食管癌细胞核的DNA断裂片段较处理前增多;琼脂凝胶电泳显示一片模糊的弥散带。结论10-1g/ml的杏多糖与阿魏蘑菇醇提物可促进食管癌细胞抑癌基因Bax的表达,并促进细胞的凋亡。
Objective To investigate the biological mechanism of apricot polysaccharides and PleurotasSapidus in esophageal cancer(Ec)cell line Eca9706.Methods The indexes of cell survival,the protein expression of Bcl-2/Bax gene and apoptosis were detected in Eca 9706 using immunochemical and molecular biology assay after treatment of apricot polysaccharides and Pleurotas Sapidus at concentration of 10^-1 g/ml.Results The concentration of 10^-1 g/ml apricot polysaccharides and Pleurotas Sapidus showed that immunochemical staining Bax gene was overexpressed.Moreover,Bcl-2 gene expression indicated a tendency to increase.The molecular biology result showed that DNA extracted from treated Eca9706 appeared a smear phenomenon.Conclusions The concentration of 10^-1 g/ml apricot polysaccharides and Pleurotas Sapidus could induce apoptosis of Eca9706 by activating anti oncogene Bax.
